Starmyu erzählt die Geschichte von fünf Schülern, Yuuta Hoshitani, Tooru Nayuki, Kaito Tsukigami, Kakeru Tengenji und Shuu Kuga. Sie kämpfen darum, der Ayanagi Academy beizutreten, welche eine Schule ist, die sich auf Musik spezialisiert hat. Sie müssen von der Star Frame Class akzeptiert werden, welche direkt von Mitgliedern der Kaou-tai unterrichtet wird. Die Kaou-tai stehen an der Spitze der Rangliste, als die Talentiertesten aus der Musikabteilung. Glücklicherweise werden sie von Itsuki Ootori entdeckt, einem Mitglied von Kaou-tai.
